Aggregation of amyloid beta in brain triggers a slowly progressing sequence of mildly sy naptotoxic and neurotoxic activities\, preceding the development of clinic al dementia by more than 20 years. Cellular reactions to this chronic toxi city include synaptic dysfunction\, tau pathology in neurons with the form ation of neurofibrillary tangles and glial cell responses. Collectively\, these pathogenic activities result in chronic neurodegeneration followed b y progressive decline of global cognitive functions. As an initiator of th is chronic cascade of degenerative activities\, amyloid beta is a genetica lly validated target for drug development throughout industry. We have dis covered\, and licensed to Biogen\, aducanumab\, the first amyloid beta-rem oving antibody receiving accelerated approval by the U.S. FDA. Aducanumab removes brain amyloid beta in a dose- and time-dependent fashion\, the bio logical basis for its FDA approval. Aducanumab is derived from human antib ody-encoding memory-B cells.
